'Promiscuous' enzymes still prevalent in metabolism: Challenges fundamental notion of enzyme specificity and efficiency
ScienceDaily (Aug. 30, 2012) ? Open an undergraduate biochemistry textbook and you will learn that enzymes are highly efficient and specific in catalyzing chemical reactions in living organisms, and that they evolved to this state from their "sloppy" and "promiscuous" ancestors to allow cells to grow more efficiently. This fundamental paradigm is being challenged in a new study by bioengineers at the University of California, San Diego, who reported in the journal Science what a few enzymologists have suspected for years: many enzymes are still pretty sloppy and promiscuous, catalyzing multiple chemical reactions in living cells, for reasons that were previously not well understood.
In this study, the research team, led by Bernhard Palsson, Galetti Professor of Bioengineering at the UC San Diego Jacobs School of Engineering, brought together decades of work on the behavior of individual enzymes to produce a genome-scale model of E. coli metabolism and report that at least 37 percent of its enzymes catalyze multiple metabolic reactions that occur in an actively growing cell.
"We've been able to stitch all of the enzymes together into one giant model, giving us a holistic view of what has been driving the evolution of enzymes and found that it isn't quite what we've thought it to be," said Palsson.
When organisms evolve, it is the genes or proteins that change. Therefore, gene and protein evolution has classically been studied one gene at a time. However in this work, Palsson and his colleagues, introduce an important paradigm shift by demonstrating that the evolution of individual proteins and enzymes is influenced by the function of all of the other enzymes in an organism, and how they all work together to support the growth rate of the cell.
Using a whole-cell model of metabolism, the research team found that the more essential an enzyme is to the growth of the cell, the more efficient it needs to be; meanwhile, enzymes that only weakly contribute to cell growth can remain 'sloppy.' The study found three major reasons why some enzymes have evolved to be so efficient, while others have not:
Enzymes that are used more extensively by the organism need to be more efficient to avoid waste. To increase efficiency, they evolve to catalyze one specific metabolic reaction. When enzymes are responsible for catalyzing reactions that are necessary for cell growth and survival, they are specific in order to avoid interference from molecules that are not needed for cell growth and survival.
Since organisms have to adapt to dynamic and noisy environments, they sometimes need to have careful control of certain enzyme activities in order to avoid wasting energy and prepare for anticipated nutrient changes. Evolving higher specificity makes these enzymes easier to control.
"Our study found that the functions of promiscuous enzymes are still used in growing cells, but the sloppiness of these enzymes is not detrimental to growth. They are much less sensitive to changes in the environment and not as necessary for efficient cell growth," said Nathan Lewis, who earned a Ph.D. in bioengineering at the Jacobs School in March and is now a postdoctoral fellow at Harvard Medical School.
This study is also a triumph in the emerging field of systems biology, which leverages the power of high-performance computing and an enormous amount of available data from the life sciences to simulate activities such as the rates of reactions that break down nutrients to make energy and new cell parts. "This study sheds light on the vast number of promiscuous enzymes in living organisms and shifts the paradigm of research in biochemistry to a holistic level," said Lewis. "The insights found in our work also clearly show that fine-grained knowledge can be obtained about individual proteins while using large-scale models." This concept will yield immediate and more distant results.
"Our team's findings could also inform other research efforts into which enzymes require further study for overlooked promiscuous activities," said Hojung Nam, a postdoctoral researcher in Palsson's lab. "Besides testing and characterizing more enzymes for potential promiscuous activities, enzyme promiscuity could have far-reaching impacts as scientists try to understand how unexpected promiscuous activities of enzymes contribute to diseases such as leukemia and brain tumors," said Nam.

The show, curiously dubbed Pro Football Talk, debuts on Tuesday, September 4.? Hosted by the highly competent and funny and engaging Erik Kuselias and by an Internet hack who is the exact opposite of those three traits, the show will bring the PFT temperament and attitude to the small screen, with help from NBC talent like Rodney Harrison, Hines Ward, Doug Flutie, Ross Tucker, Amani Toomer, and Peter King.
The powers-that-be tell me they?re sending out an official press release soon.? They wanted me to say something for it.? Here?s the best I could muster:? ?When we started the website, the goal was to create a place I?d want to visit as a football fan.? While the men usually appear in the body. You should be wary when looking at such signs of fungal disease but black. Black spot will usually grow bigger is brown, reddish brown, or black permanent.
Those are some signs you should look for when looking at existing spots or warts on your skin. Look closely and be alert. Knowing skin cancer early can be very helpful in the healing process. Do not forget to continue to protect your skin.

How Bain Capital execs lower their taxes - The ... - Fortune Finance
FORTUNE -- There has been lots of talk over the past few days about how Bain Capital executives have used management fee waivers?to effectively lower their tax payments (a tactic that is not unique to Bain). Some academics have argued that such waivers are an illegal dodge, while private equity tax attorneys I've spoken with call it "aggressive but accepted by the IRS."
Here is the basic structure: Bain officially charges 2% management fees to investors in its private equity funds. The idea is to cover overhead, such as salaries, office leases, electric bills, etc. ?But Bain has lots of other business lines (venture capital funds, hedge funds, etc.) that generate sufficient cash flow, so it "waives" the PE fund management fees. Well, sort of. Limited partners still pay the 2%, only it flows directly into the fund, as part of Bain's general partner commitment (which is between 10% and 15% of Bain's total PE fund capital, an unusually high percentage).
By doing so, Bain partners don't pay ordinary income taxes on their management fees. Instead, they pay at capital gains rates if/when the deals generate profit (because it's now considered carried interest). It appears that the partners irrevocably waive their rights to the returns if the deal returns less than par, although I don't have access to the LPAs that would spell it out more exactly. That said, sources do tell me that Bain partners cannot take waiver-related capital loss on failed transactions.
Sources sympathetic to such structures cite an IRS ruling (93-27) that specifically seems to allow such waivers, under a notion of partnership risk. Critics say that rule is being perverted, because the risk of loss is so low.
I'm not well-versed enough in IRS minutia to render judgment, but do feel that such structures should be banned going forward. Management fees are a fee for service. As such, they should be taxed as ordinary income. If Bain wants to eliminate management fees altogether, then move to a 0-and-32 structure.
To be sure, I have no problem with fund managers pushing some of their fees into funds. I think it's a good thing, as it's a tighter alignment of interests. But I don't see why their capital should be cheaper than that of the high-net-worth investors in their funds (who paid taxes on their pre-commitment monies). Doing so, in fact, depresses that alignment.
Tax law is a funny thing, because certain?strategies aren't black and white. They're subject to interpretation. When I asked a PE tax attorney if what Bain is doing is allowed, she told me that "allowed" and "not allowed" isn't always the right way to think about it. Okay, understood. But some things are explicitly permitted (treating carried interest as capital gains, for example) and other things are explicitly prohibited (taking deductions for work expenses for which I was reimbursed). So let's just add this one to the latter list.

Can Curiosity Mars mission inspire like Apollo?
LOS ANGELES (AP) ? Neil Armstrong inspired millions with his moonwalk. Can a feisty robotic rover exploring Mars do the same for another generation? With manned missions beyond the International Space Station on hold, the spotlight has turned on machines.
While it did not rise to Armstrong's "one small step for man, one giant leap for mankind," interest was so high in the rover Curiosity's "seven minutes of terror" approach to the red planet earlier this month that NASA's website crashed after receiving nearly 2 billion hits. The rover last week beamed home photographs of its first wheel tracks on the Martian soil since its daredevil landing
"There's something exciting about reaching another place in the solar system. If you think about the kind of interest the landing of Curiosity had, you get a sense of that," said Smithsonian Institution space curator Roger Launius. It wasn't on the same level as Armstrong's feat, "but it was pretty darn exciting," he said.
When Armstrong, then fellow astronaut Buzz Aldrin, stepped on the moon on July 20, 1969, an estimated 600 million people watched and listened. "Virtually the entire world took that memorable journey with us," recalled Buzz Aldrin after Armstrong's death Saturday.
Early in the Space Age, the Mercury, Gemini and Apollo astronauts were the public face of NASA's space endeavor while the unmanned lunar missions that paved the way were in the shadows. The public craved adventure and the manned missions delivered. Aiming for the moon was new and exciting ? not to mention dangerous ? and the U.S. was locked in a Cold War space race with the Soviets.
Next, the space shuttle ferried a new crop of astronauts to low-Earth orbit, but after three decades of service, it became routine. And the Cold War thawed with the Russians and Americans cooperating on the Russian space station Mir and the International Space Station.
With the space shuttle fleet retired, the space station is all that's left. Its crew of six for the most part quietly goes about doing its job about 250 miles above the Earth. President Barack Obama nixed plans for returning astronauts to the moon in favor of landing on an asteroid and eventually Mars.
These days, space exploration is carried out by robotic spacecraft ? commanded by human handlers on Earth. Advances in technology have allowed unmanned spacecraft to go farther and peer deeper, with craft circling Mercury, Saturn, asteroid Vesta and others headed for Jupiter and dwarf planet Pluto. The twin Voyager craft are still going strong at the fringes of the solar system 35 years after their launch in 1977.
American University space policy analyst Howard McCurdy said today's generation of explorers was raised on technology and tends to get more jazzed about delivering a car-size rover to Mars.
"Robotic exploration has taken more of a center stage," he said. "It gets more publicity now than the International Space Station."
When the first Mars rover Sojourner landed in 1997, science fiction writer Arthur C. Clarke rephrased Armstrong's famous line and said the event was "one small step for the rover."
Three other rovers have followed including Curiosity, which landed Aug. 5 by executing an intricate routine that ended with it being lowered by cables to the surface. Curiosity's acrobatics proved so popular that its Twitter followers surged from 120,000 the eve of landing to more than a million (the tweets are being written by the public affairs office at the NASA Jet Propulsion Laboratory, which manages the $2.5 billion mission.)
Curiosity chief scientist John Grotzinger said Monday the wheel prints on Mars may turn out to be an iconic image just like those first boot prints on the lunar surface.
"Instead of a human, it's a robot pretty much doing the same thing," he said.
Henry Lambright, a professor of public policy and space scholar at Syracuse University, said while Curiosity is inspiring, the world still needs to send humans beyond low-Earth orbit.
"It can't inspire to the degree that Apollo did because a robot can't inspire the way a man can," Lambright said.
On Monday, NASA played a recording from Administrator Charles Bolden that had been sent up to the rover on Mars and relayed back to Earth. In it, he thanked scientists and engineers for their achievement.
David Lavery of NASA headquarters said the hope is that someone will be inspired by Bolden's message and become the first human to stand on Mars.
"Like the great Neil Armstrong, they'll be able to speak aloud ? the first person at that point, of the next giant leap in human exploration," he said.

ICANN will then sift through the applications and comments to decide which domain names to approve. The organization hopes to begin rolling out new domain names in 2013.
The National Restaurant Association is part of the Coalition for Responsible Internet Domain Oversight, an effort by more than 160 business groups and companies to urge a deliberative process as domain names are expanded. CRIDO is concerned about confusion for Internet users as well as the possibility of huge new costs if businesses are forced to register new domains to protect their trademarks and brands.

Still, he admitted that the pair's DNA is in musical films like "Chicago," "Footloose" and "Hairspray," and in Broadway musicals like the recent revivals of "How to Succeed in Business Without Really Trying" and "Promises, Promises."
It's probably safe, then, to figure that their musical theater background will influence what they put on the stage of the Dolby Theatre. And while it's not fair to saddle the new guys with the hits and misses of their predecessors, it might be instructive to take a look at what other musical vets did with the Oscar stage when they had a shot at it.

He also loosened the dress code, had a rock band play while dancers wearing the nominated costumes danced the frug and the jerk, and moved the Best Actor award to the middle of the show.
Upon his arrival at the Governors Ball, guests greeted Champion with a standing ovation.
1970: ROBERT WISE
Two years after Champion's show, "West Side Story" and "The Sound of Music" director - and future AMPAS president - Wise took the group-hosting concept to its extreme. Abandoning the idea of a host entirely, Robert Wise used 32 different "friends of Oscar" - or, as we might call them these days, presenters.
Wise also insisted that everybody appearing on the show be a true movie star - which meant, for example, that the Carpenters had to turn over the performance of their nominated song "For All We Know" to Petula Clark, because she had an onscreen career and they didn't.
To a degree, Wise was hamstrung by the fact that only one of the acting winners showed up and that all the suspense revolved around whether the Best Actor award would go to George C. Scott, who had said he didn't want to be nominated for "Patton" and wouldn't accept if it won. He did win, and he didn't accept.
Afterward, according to the book "Inside Oscar," Daily Variety summed it up: "At 43, Oscar looked tired."
1985: STANLEY DONEN
In 1998, director Stanley Donen gave one of the most delightful acceptance speeches in Oscar history, singing "Cheek to Cheek" while dancing with the honorary Oscar with which he'd just been presented. But a dozen years earlier, the director of such classic musicals as "Singing in the Rain," "On the Town," "Seven Brides for Seven Brothers" and "Damn Yankees" had produced the Oscars, with mixed results.
His show began with an elaborate and unfortunate opening number featuring film clips from "Flying Down to Rio" interspersed with shots of actress Teri Garr piloting an airplane and then climbing onto the wing with a chorus line of dancing girls.
"After that, things improved considerably," the Academy's official history understates - and, in fact, the rest of the show, which included Howard Keel serenading a bevy of leading ladies from MGM musicals, was for the most part enthusiastically received. Despite the opening number, the New York Times called Donen's work "the best Oscar show in years, maybe ever."
1988: ALLAN CARR
You might have heard about this one. To spice up the Oscars at the end of the '80s, the Academy turned to Allan Carr, who had worked on "Saturday Night Fever," co-produced and written "Grease," produced the Village People musical "Can't Stop the Music" and produced the Broadway musical version of "La Cage aux Folles."
Carr immediately proclaimed that he was going to stage the most fabulous Oscars ever. Instead, he staged the most infamous.
His show opened with a monstrous 11-minute musical extravaganza whose most memorable moment came when Rob Lowe butchered a rewritten version of "Proud Mary" with an actress dressed as Snow White - a character for which Carr had forgotten to get clearance. A later musical number, "I Wanna Be an Oscar Winner," was almost as long and just as bad.
Disney sued over the Snow White fiasco, the critics trashed the show, and many said that Carr never recovered from his very public failure.
1996: QUINCY JONES
Quincy Jones had made a bigger mark in the music industry, where he's set the record for most Grammy nominations with 79, than he had in movies when the Academy asked him to produce the Oscars in 1996. But he'd also produced "The Color Purple," written music for films and won the Jean Hersholt Humanitarian Award, and he enthusiastically took the job with co-producer David Salzman, saying again and again that he was going to change everything about the Oscars.
He didn't quite accomplish that, and during rehearsals his Oscars appeared to be an impending train wreck, chaotic and disorganized.
But on the night of the show it all came together - and once you got past a silly fashion-model presentation of the costume awards, Jones' Oscars had energy (Stomp), music (Bruce Springsteen), eloquence (Emma Thompson's acceptance speech) and real emotion (Christopher Reeve's first public appearance since being paralyzed in a riding accident).
2008: BILL CONDON & LAURENCE MARK
Writer-director Bill Condon, who tackled the Oscars with producer Laurence Mark, got the Academy job after directing one musical, "Dreamgirls," and writing another, Best Picture winner "Chicago." But he and Mark showed a theatrical flair in their Oscar show, with frequent Broadway leading man Hugh Jackman winning raves for his job as host.
If you can get past the incongruity of Jackman singing an opening number about a downsized, lower-budgeted Oscars while standing beneath a million-dollar crystal Swarovski curtain, the Condon and Mark show was the freshest and best of the recent Oscar shows, nicely re-staging the Best Actor and Actress presentations and bringing a shot of adrenaline and intimacy to the entire enterprise.
2009: ADAM SHANKMAN (WITH BILL MECHANIC)
The year after Condon and Mark had their turn, "Hairspray" and "Rock of Ages" director Adam Shankman, who'd begun his career as a choreographer, teamed up with producer Bill Mechanic for an Oscars that was not exactly as successful as its predecessor.
"It wasn't the worst Oscars ever, but it may have been the most disappointing," wrote Joe Adalian at TheWrap, complaining that the show was conventional and tired.
The Shankman/Mechanic show, which did away with performances of the nominated songs for the first time since the Allan Carr Oscars, included Neil Patrick Harris singing a lavish opening number before handing the reins to co-hosts Steve Martin and Alec Baldwin.
Shankman also hired 69 dancers, many from the TV show "So You Think You Can Dance," on which he served as a judge - but their use never seemed anywhere near as exciting and revolutionary as he'd promised.
Shankman's and Mechanic's biggest innovation - changing the phrase "and the Oscar goes to... " back to the edgier "and the winner is... " - was abandoned the following year.

Romney promises a countervision of "growth and jobs and economic vitality."
There are no shades of gray. Beyond the usual election-year distortions and exaggerations, a yawning issues gulf separates the two candidates.
Romney sharpened the distinctions by selecting Wisconsin Republican Rep. Paul Ryan as his running mate, a choice that cheered the GOP's conservative base.
The differences will be on display over the next two weeks as Romney, and then Obama, host their national party conventions.

Obama sees government as the catalyst for economic growth, a safety net for the neediest, a protector against free-market excesses and a safeguard for the environment. "The old rule-of-thumb was that to win the nomination you first win your base and then move to the middle. Both Obama and Romney are being forced to play it closer to their bases than in the past," said presidential scholar Stephen Hess at the Brookings Institution think tank. By now, both candidates should have begun a migration to the center to attract crucial independent voters. "But that's not happening," Hess said.

Romney governed Massachusetts as a political moderate and reached across the aisle to majority-party Democrats. Some analysts suggest he may again govern from the middle if elected.
But so far Romney hasn't strayed much from conservative dogma, while Obama has abandoned serious efforts to cut deals with Republicans.
If Romney really is a moderate at heart, "he hasn't made it any easier for himself in the campaign so far," said William Galston, a top domestic policy adviser to then-President Bill Clinton.
GOP consultant Rich Galen suggests the sharp Obama-Romney divide reflects divisions across America but more immediately echoes the harsh, unforgiving partisan rhetoric in Congress.
"It's becoming much more like the British system where, no matter what the government does, the opposition hates it. It's never a good idea," Galen said. "I'm not sure it's the best possible course for our republic."
Romney survived a brutal GOP primary season only by repeatedly fending off charges by rivals from his own party that he was too moderate. And Obama moved closer to his Democratic base as the recovery slowed and as Republicans made his defeat a top priority.
Norman Ornstein of the American Enterprise Institute said Romney may have been permanently altered by the rings of fire he had to jump through to prove he was conservative enough to carry the GOP banner.
"If you take only the public statements and the promises that have been made, the choice is a stark one," Ornstein said, "starker than we've seen before."

Judge OKs stem cell cure for 2-year-old girl
MILAN (AP) ? Doctors are preparing an emergency one-off stem cell treatment for 2-year-old Venetian girl suffering a severe muscular disease after a judge revoked an order blocking the cure.
Brescia hospital officials said Thursday the treatment for Celeste Carrer would begin within days. Carrer suffers from spinal muscular atrophy, which has a life expectancy of about two years. Her family reported marked improvement after beginning the experimental treatment this winter.
But the stem cell lab was shut down in May after Italy's drug agency determined it was not hygienic and had violated procedures. A judge in Venice has allowed one treatment pending a final ruling, expected next week, on the family's appeal.
Besides reactivating the lab, doctors must determine if the stem cells from the girl's mother remain viable.
